目的探讨职业砷接触工人mi RNA生成和成熟相关m RNA表达变化规律,深入认识砷的致癌机制。方法整群随机抽取正在进行砷冶炼的工人43名作为高暴露组,已经停止砷冶炼85 d的工人36名作为低暴露组,对照组24人。用实时荧光定量PCR(RT-PCR)检测外周血mi RNA生成和成熟相关m RNA表达情况,采用氢化物发生原子吸收分光光度法检测尿砷含量。尿砷浓度和相关m RNA的-△△Ct值组间比较采用t检验。检验水准α=0.05。结果高暴露组尿砷为(1738.62±1361.62)μg/g肌酐,低暴露组为(561.20±451.50)μg/g肌酐,对照组为(17.63±11.07)μg/g肌酐,各组间差异具有统计学意义(P<0.01)。高暴露组外周血mi RNA生成和成熟相关m RNA Lin28、Lin28b、Exportin-5、Trbp、Dicer、Ago2 RT-PCR扩增-△△Ct与低暴露组和对照组间差异均具有统计学意义(P<0.01),而低暴露组与对照组间差异均无统计学意义(P>0.05)。结论砷冶炼工人机体内可能存在大量mi RNA生成和成熟相关m RNA表达增加。此过程是可逆的,砷中毒患者早诊、早治非常重要。
OBJECTIVE: To investigate the changes of m RNA expression related to mi RNA production and maturation in workers exposed to arsenic, and to understand the carcinogenic mechanism of arsenic. Methods A total of 43 workers who were arsenic smelting were selected randomly as the high exposure group and 36 workers who stopped smelting for 85 days as the low exposure group and 24 control subjects. Real-time quantitative PCR (RT-PCR) was used to detect the mRNA expression of miRNA in peripheral blood and maturation, and the content of arsenic in urine was detected by hydride generation atomic absorption spectrophotometry. Arsenic concentrations in urine and related - ΔΔCt values of m RNA were compared between groups using t test. Test level α = 0.05. Results The urinary arsenic in high exposure group was (1738.62 ± 1361.62) μg / g creatinine, the low exposure group was (561.20 ± 451.50) μg / g creatinine, and the control group was (17.63 ± 11.07) μg / g creatinine. Significance (P <0.01). The differences of miRNA production and maturation in peripheral blood of high exposure group were statistically significant (P <0.05) with those of low exposure group and control group (Lin28, Lin28b, Exportin-5, Trbp, Dicer, Ago2 RT- P <0.01), but there was no significant difference between low exposure group and control group (P> 0.05). Conclusion Arsenic smelters may have a large number of miRNAs that are involved in miRNA production and maturation. This process is reversible, arsenic poisoning early diagnosis, early treatment is very important.
