目的探讨小鼠急性心肌梗死模型建立及左心室重构评价。方法取40只C57B/L6雄性小鼠,30只结扎冠状动脉(冠脉)左前降支,建立急性心肌梗死模型(A组),10只予假手术为对照(B组)。术前1d和术后4周心脏超声评价左心室舒张末内径(LVEDD)和左室射血分数(LVEF)。术后4周测量小鼠心脏重量/体重(HW/BW)比值,取心脏标本做Masson染色测量梗死纤维化面积,WGA染色测量小鼠心肌细胞横截面积。结果 A组心肌梗死模型建立成功率为73.3%(22/30)。A组小鼠术后4周LVEDD、心肌细胞横截面积和HW/BW比值均大于B组[(4.45±0.57)mm vs.(3.44±0.36)mm、(746.60±74.48)μm2 vs.(296.20±37.21)μm2和0.0065±0.0010vs.0.0054±0.0004](P<0.05)。A组小鼠术后4周LVEF低于B组[(46.95±6.72)%vs.(73.79±6.56)%](P<0.05)。A组平均梗死后纤维化面积为(48±11)%。结论采用结扎冠脉前降支可成功建立小鼠急性心肌梗死模型。LVEDD、LVEF HW/BW比值等可用于综合评价梗死后心肌重构程度。
Objective To investigate the establishment of acute myocardial infarction model in mice and the evaluation of left ventricular remodeling. Methods Forty C57B / L6 male mice and 30 left anterior descending coronary arteries were ligated to establish acute myocardial infarction model (A group) and 10 sham operation group (B group). Left ventricular end diastolic diameter (LVEDD) and left ventricular ejection fraction (LVEF) were assessed by echocardiography 1 day before surgery and 4 weeks after surgery. The ratio of heart weight to body weight (HW / BW) was measured 4 weeks after operation. The area of infarction fibrosis was measured by Masson staining and the myocardial cell cross - section was measured by WGA staining. Results The success rate of myocardial infarction model in group A was 73.3% (22/30). The LVEDD, myocardial cell cross-sectional area and HW / BW ratio in group A at 4 weeks after operation were significantly higher than those in group B [(4.45 ± 0.57) mm vs. (3.44 ± 0.36) mm, (746.60 ± 74.48) μm2 vs. ± 37.21) μm 2 and 0.0065 ± 0.0010 vs. 0.005 ± 0.0004] (P <0.05). The LVEF in group A was lower than that in group B at 4 weeks (46.95 ± 6.72% vs 73.79 ± 6.56%, P <0.05). The mean infarct fibrosis area in group A was (48 ± 11)%. Conclusion The model of acute myocardial infarction in mice can be successfully established by ligating the anterior descending coronary artery. LVEDD, LVEF HW / BW ratio can be used to evaluate the degree of myocardial remodeling after infarction.
