目的:探讨血管内皮生长因子121(VEGF 121)在小鼠囊膜组织和体外培养的新生小牛晶体上皮细胞中的表达情况。方法:分别完整的取下11个月龄的20只DBA/2J高眼压模型小鼠和20只正常BL6小鼠的晶状体囊膜组织,提取蛋白后,利用Western blot方法检测晶状体囊膜组织VEGF 121的表达。体外培养牛眼晶体上皮细胞,利用免疫细胞化学染色SP法检测晶体上皮细胞中VEGF121的表达。结果:Western blot方法显示两种小鼠囊膜均有VEGF121的表达,且两种小鼠的表达无明显差异。牛眼晶体上皮细胞也有VEGF121阳性颗粒的表达,阴性对照中未见阳性染色。结论:两种小鼠晶状体囊膜组织和体外培养的牛眼晶体上皮细胞均有VEGF 121的表达,表明晶状体中产生的VEGF 121可能参与小鼠胚胎发育和眼内某些新生血管性疾病的发生。
OBJECTIVE: To investigate the expression of vascular endothelial growth factor 121 (VEGF 121) in cultured mouse calvaria tissue and cultured neonatal calf epithelial cells. Methods: The lens capsule of 20 mice with DBA / 2J ocular hypertension and 20 normal BL6 mice of 11 months old were completely removed. After extraction of protein, Western blot was used to detect the expression of VEGF 121 expression. Bovine lens epithelial cells were cultured in vitro, and the expression of VEGF121 in lens epithelial cells was detected by immunocytochemical stain SP method. Results: The results of Western blot showed that the expression of VEGF121 was found in both mouse capsular membranes, and there was no significant difference between the two mice. Bovine eye lens epithelial cells also have VEGF121-positive particles, no positive staining in the negative control. CONCLUSION: The expression of VEGF121 in both lens capsule and bovine lens epithelial cells cultured in vitro suggests that VEGF 121 in lens may be involved in mouse embryonic development and certain intraocular neovascular diseases .
