目的探讨中波紫外线(UVB)对人胃癌细胞SGC-7901的损伤,建立UVB损伤人胃癌细胞的模型,为防护紫外线损伤提供技术支持。方法取体外培养的人胃癌细胞SGC-7901,用0(对照组)、6、12、18 m J/cm2的UVB照射后继续培养5h,以MTT法检测细胞的增殖活性,在光学显微镜下观察细胞的形态学改变,用Annexin-FITC细胞凋亡试剂盒检测细胞凋亡。结果 UVB照射可使SGC-7901细胞增殖活性降低,且呈剂量效应关系(F=864.302,P<0.01);形态学上可见细胞形态肿胀变圆,失去与邻近的细胞的连接;紫外线照射可诱导SGC-7901细胞发生凋亡。结论 UVB可损伤SGC-7901细胞,降低其增殖活性,诱导其发生细胞凋亡,并且呈明显的剂量效应关系。
Objective To explore the damage of human gastric cancer cell line SGC-7901 by UVB and establish the model of UVB-induced human gastric cancer cell line, in order to provide technical support for the protection of ultraviolet light. Methods SGC-7901 human gastric cancer cell line SGC-7901 was cultured in vitro and incubated with 0 (control) 6,12,18 m J / cm2 UVB for 5 hours. The proliferative activity of SGC-7901 cells was detected by MTT assay. Morphological changes of cells were detected by Annexin-FITC apoptosis kit. Results UVB irradiation reduced the proliferation of SGC-7901 cells in a dose-dependent manner (F = 864.302, P <0.01). Morphologically, cell morphology became round and the adjacent cells were lost, and UV irradiation could induce SGC-7901 cells undergo apoptosis. Conclusion UVB can damage SGC-7901 cells, reduce their proliferative activity and induce apoptosis of SGC-7901 cells, and show a dose-dependent effect.
