目的:探讨醛固酮(ALD)对大鼠肾小球系膜细胞纤溶酶原激活物抑制物1(PAI-1)基因表达的影响。方法:(1)以不同浓度的ALD(10-11、10-10、10-9、10-8、10-7mol/L)刺激肾小球系膜细胞72 h后,用半定量RT-PCR法检测该细胞中PAI-1 mRNA的表达。(2)以10-7mol/L的ALD刺激肾小球系膜细胞不同时间(12、24、48、72 h)后,用半定量RT-PCR法检测该细胞中PAI-1 mRNA的表达。结果:半定量RT-PCR结果显示,ALD促进培养的大鼠肾小球系膜细胞PAI-1mRNA的表达,且具有浓度依赖性和时间依赖性的特点。结论:ALD促进大鼠肾小球系膜细胞PAI-1 mRNA的表达,从而抑制细胞外基质的降解,促进肾小球疾病进展。
Objective: To investigate the effect of aldosterone (ALD) on the expression of plasminogen activator inhibitor 1 (PAI-1) in rat mesangial cells. Methods: (1) The mesangial cells were stimulated with different concentrations of ALD (10-11, 10-10, 10-9, 10-8, 10-7mol / L) PAI-1 mRNA expression was detected in this cell line. (2) The mesangial cells were stimulated with 10-7mol / L ALD for different time (12,24,48,72 h), and the expression of PAI-1 mRNA was detected by semi-quantitative RT-PCR. Results: The results of semi-quantitative RT-PCR showed that ALD promoted the expression of PAI-1mRNA in cultured rat mesangial cells in a concentration-dependent and time-dependent manner. Conclusion: ALD can promote the expression of PAI-1 mRNA in rat mesangial cells and inhibit the degradation of extracellular matrix and promote the progression of glomerular diseases.
