目的:观察活血潜阳方对高血压大鼠内皮祖细胞的影响。方法:正常血压WKY大鼠10只,自发性高血压大鼠(SHR)30只(随机分为模型组、活血潜阳方组、代文组),WKY组、模型组以生理盐水2 ml/d灌胃,活血潜阳方组予活血潜阳方(0.96 g·kg-1·d-1)、代文组予代文溶液(20 mg·kg-1·d-1)灌胃,共干预8周。在实验前及灌胃4、8周时用尾动脉容积法测定血压,HE染色观察胸主动脉内皮细胞层连续性,并测定循环内皮祖细胞数量及其功能。结果:活血潜阳方组、代文组血压与模型组比较差异有统计学意义(P<0.01),代文组降压效果优于活血潜阳方组(P<0.01)。模型组大鼠胸主动脉内皮层受损最为显著,而活血潜阳方组和代文组大鼠相同部位血管内皮均获一定程度修复,且其循环内皮祖细胞数量及增殖、黏附和迁移能力显著高于模型组(P<0.01,P<0.05)。结论:SHR大鼠胸主动脉内皮细胞完整性严重破坏,循环内皮祖细胞数量减少、功能下降;活血潜阳方可提高循环内皮祖细胞数量和功能。
Objective: To observe the effect of Huoxue Qianyang Recipe on endothelial progenitor cells in spontaneously hypertensive rats. Methods: 10 normal WKY rats, 30 spontaneously hypertensive rats (SHR) were randomly divided into model group, Huoxueqianyang group and Daiwen group, WKY group. The model rats were treated with normal saline (2 ml / The rats in the Xueyang Qianyang prescription group were treated with Huoxueqianyang prescription (0.96 g · kg -1 · d -1), and the Daiwen injection was administered with 20 mg · kg -1 · d -1 Intervention for 8 weeks. Blood pressure was measured by caudate arterial volume method before and 4 and 8 weeks after gavage. The continuity of thoracic aorta endothelial cells was observed by HE staining. The number of circulating endothelial progenitor cells and its function were determined. Results: There were significant differences in the blood pressure between the Huoyang Qiyangfang group and the Daiwen group and the model group (P <0.01). The antihypertensive effect of the Daiwen group was better than that of Huoxueqianyang group (P <0.01). In model group, thoracic aorta endothelial cell damage was the most significant, and the same part of Huoxue Qianyang and Daiwen rats had a certain extent of vascular endothelial repair, and the number of circulating endothelial progenitor cells and proliferation, adhesion and migration ability Significantly higher than the model group (P <0.01, P <0.05). Conclusion: The integrity of endothelial cells of thoracic aorta in SHR rats is severely damaged, the number of circulating endothelial progenitor cells is decreased and the function is decreased. Huoxue Qianyang can increase the number and function of circulating EPCs.
