目的 我们首次对雷氏大疣蛛 (Macrotheleraveni)蛛毒对人肝癌SMMC-7721细胞和人胃癌BGC-823增殖的抑制作用进行研究,体外实验进一步探讨其作用机理。方法 采用MTT法测定雷氏大疣蛛蛛毒对SMM-C7721和BGC- 823细胞增殖作用的影响;利用流式细胞光度术(FCM)探讨雷氏大疣蛛蛛毒对细胞凋亡率的影响。结果MTT方法表明雷氏大疣蛛蛛毒对两种细胞增殖有较强的抑制作用(P<0. 05),时效和量效关系良好。流式细胞仪检测发现经过雷氏大疣蛛蛛毒 (10, 20, 40μg/mL)作用下 24小时后两种细胞凋亡率增加。结论 雷氏大疣蛛蛛毒可以抑制两种细胞的增殖,其可能的作用机理是在一定的浓度下诱导肿瘤细胞凋亡。
Objective To investigate the inhibitory effect of Macrotheleraveni spider venom on the proliferation of human hepatoma SMMC-7721 cells and human gastric cancer cell line BGC-823 for the first time, and to explore its mechanism in vitro. Methods MTT assay was used to determine the effects of spider venom on the proliferation of SMM-C7721 and BGC-823 cells. Flow cytometry (FCM) was used to investigate the effects of spider venom on the apoptosis rate. Results The results of MTT assay showed that the spider venom was resistant to both cell proliferation (P <0.05), and the relationship between aging and dose-effect was good. Flow cytometry showed that the apoptosis rate of the two cells increased after 24 hours by the action of 10, 20, 40μg / mL. Conclusion The spider venom of Reicularia spp. Can inhibit the proliferation of two kinds of cells, and its possible mechanism is to induce tumor cell apoptosis under a certain concentration.
