目的:观察过表达人全长tau蛋白对原代海马神经元胞内钙信号和神经元形态的影响及其机制。方法:通过钙成像观察过表达人全长tau蛋白的神经元胞内钙离子浓度的变化。采用免疫荧光染色观察过表达tau蛋白的神经元树突及树突棘的变化。免疫印迹观察过表达tau蛋白的神经元内钙神经素的蛋白变化。结果:过表达人全长tau蛋白的神经元胞内基础钙水平
OBJECTIVE: To investigate the effect of overexpression of human full-length tau on intracellular calcium signaling and neuronal morphology in primary hippocampal neurons and its mechanism. Methods: Changes of intracellular calcium concentration in neurons overexpressing human full-length tau were observed by calcium imaging. Immunofluorescence staining was used to observe the changes of dendrites and dendritic spines in neurons overexpressing tau. Immunoblotting was used to observe changes in protein content of intracellular calcineurin over-expressing tau protein. RESULTS: Intracellular basic calcium levels were overexpressed in human full-length tau protein
