目的研究热休克蛋白70(Hsp70)和肿瘤抗凋亡蛋白bcl-2在涎腺腺样囊性癌(Salivary adenoid cystic carcino-ma,SACC)中的表达,并探讨与SACC发生、发展之间的关系及临床意义。方法采用免疫组化通用型二步法检测36例SACC中Hsp70及bcl-2的表达,并选用10例多形性腺瘤(plemorphic adenomas,PAS)作对照。结果Hsp70表达为细胞核及胞浆中棕黄色染色,在SACC中的阳性表达为86.1%,与对照组PAS比较差异显著(P<0.01)。而bcl-2的表达为细胞浆着色,染色呈棕黄色或棕褐色,在SACC中的阳性表达为75%,与对照组PAS比较差异显著(P<0.05)。Hsp70表达与肿瘤临床分期、组织学分型及远处转移呈正相关(P<0.05),但与局部复发无相关性(P>0.05)。bcl-2表达与临床分期呈正相关(P<0.05),但与组织学分型、局部复发及远处转移无相关性(P>0.05)。结论Hsp70可作为判断患者预后的指标。Hsp70与bcl-2在SACC中的表达,经统计学分析认为无相关性(P>0.05),Hsp70与bcl-2并未见有显著的协同关系,二者可能通过不同的途径抑制细胞的凋亡。
Objective To investigate the expression of heat shock protein 70 (Hsp70) and tumor anti-apoptotic protein bcl-2 in salivary adenoid cystic carcinomas (SACC) and to explore the relationship between the expression of Hsp70 and the development and progression of SACC Relationship and clinical significance. Methods The expression of Hsp70 and bcl-2 in 36 cases of SACC was detected by immunohistochemical general two-step method. Ten cases of plemorphic adenomas (PAS) were selected as controls. Results The expression of Hsp70 was brown-yellow in nucleus and cytoplasm. The positive expression of Hsp70 in SACC was 86.1%, which was significantly different from that in control group (P <0.01). However, the expression of bcl-2 was cytoplasm stained, stained brown or tan, and the positive expression of bcl-2 was 75% in SACC, which was significantly different from that in control group (P <0.05). Hsp70 expression was positively correlated with clinical stage, histological type and distant metastasis (P <0.05), but not with local recurrence (P> 0.05). The expression of bcl-2 was positively correlated with clinical stage (P <0.05), but not with histological type, local recurrence and distant metastasis (P> 0.05). Conclusion Hsp70 can be used as an index to judge the prognosis of patients. There was no correlation between Hsp70 and bcl-2 expression in SACC (P> 0.05), and there was no significant synergistic relationship between Hsp70 and bcl-2.
