阳离子转运载体HKT(high-affinity K~+transporter)类蛋白既是高亲和的K~+转运载体,也是一种Na~+转运体,具有Na~+和K~+转运的双重功能,对调节细胞内Na~+/K~+动态平衡起着决定性作用。胡杨长期生长在盐渍化和干旱的土壤环境,对高盐和干旱形成了极强的适应能力,是典型的耐盐抗旱植物,成为研究多年生林木抗逆适应机制的理想材料。以胡杨根系为材料,本研究克隆鉴定了一个胡杨Peu HKT1基因,该基因含有3个外显子和2个内含子;其c DNA全长为1 076 bp,包括13 bp的5’端非翻译区(5’UTR)和232 bp的3’端非翻译区(3’UTR);长831 bp的开放阅读框(open reading frame,ORF)可编码276个氨基酸;其编码蛋白含有丰富的α-螺旋,存在多个跨膜结构域,蛋白质相对分子量(MW)为31.54 k D;理论等电点(p I)9.36;实时定量PCR技术构建了Peu HKT1基因在高盐胁迫条件下的动态表达模式,探讨了该基因参与胡杨高盐胁迫响应的信号转导途径。
The high-affinity K ~ + transporter (HKT) protein is both a high-affinity K ~ + transporter and a Na ~ + transporter with dual functions of Na ~ + and K ~ + transport. Intracellular Na ~ + / K ~ + homeostasis plays a decisive role. Populus euphratica long-term growth in the salinization and arid soil environment, high salt and drought formed a strong adaptability, is a typical salt tolerance and drought resistance plants, as adaptation of perennial forest adaptation mechanism ideal material. In this study, Populus euphratica Peu HKT1 gene was cloned and identified. The gene contains three exons and two introns. The full-length c DNA is 1 076 bp, including a 13 bp 5 ’ (3’UTR) of 3’UTR and a 232 bp open reading frame (ORF) encoding 276 amino acids. The encoded protein was rich in α - helix, multiple transmembrane domains existed, the relative molecular weight (MW) of the protein was 31.54 kD and the theoretical isoelectric point (p I) was 9.36. The dynamic expression of Peu HKT1 gene under high salt stress was constructed by real-time PCR The signal transduction pathway involved in the response of Populus euphratica to salt stress was discussed.
