目的:通过观察脾虚状下大鼠oatp2a1基因表达来探讨Oatp2a1在湿浊转运中的作用。方法:32只大鼠随机分为4组,分别为正常组(6只)、正常+AA大鼠模型组(6只)、脾虚大鼠模型组(10只)、脾虚+AA大鼠模型组(10只)。采用皮下注射利血平的方法造脾虚大鼠模型,造模成功后给予相应组别大鼠马兜铃酸灌胃3天,最后采用实时荧光定量PCR方法检测各组大鼠肺、肝、肾、胃、小肠、大肠等组织中Oatp2a1的基因表达情况。结果:Oatp2a1在上述6种脏器中都有不同程度的表达。肝组织中,脾虚组大鼠模型Oatp2a1的表达量较正常组明显升高(P=0.035,P<0.05);小肠组织中,脾虚组大鼠模型中Oatp2a1的表达较正常组明显下降(P=0.004,P<0.01)。在给予马兜铃酸刺激下,小肠组织中,正常+AA组大鼠模型Oatp2a1的表达较正常组明显下降(P=0.032,P<0.05)。结论:Oatp2a1可能是机体参与湿浊运化的物质基础之一,在脾虚状态下大鼠小肠、肝组织中Oatp2a1的表达变化提示小肠、肝等脏器在脾虚状态下的湿浊转运中可能发挥重要作用,脾主运化湿浊的功能,可能不仅包括胃肠道的功能在内,而且也可能包括一部分肝脏的功能,值得进一步研究。
OBJECTIVE: To investigate the role of Oatp2a1 in wet-turbid transport by observing the expression of oatp2a1 gene in spleen-like rats. Methods: Thirty-two rats were randomly divided into four groups: normal group (6), normal + AA model group (6), spleen deficiency model group (10), spleen deficiency + AA model group (10). Sprague-Dawley rats were injected subcutaneously with reserpine, and the rats in the corresponding groups were given gavage for 3 days after successful modeling. Finally, real-time fluorescence quantitative PCR was used to detect the lung, liver and kidney , Gastric, small intestine, large intestine and other tissues Oatp2a1 gene expression. Results: Oatp2a1 had different expression levels in the above six kinds of organs. In liver tissue, the expression of Oatp2a1 in spleen deficiency group was significantly higher than that in normal group (P = 0.035, P <0.05); in small intestine, the expression of Oatp2a1 in spleen deficiency group was significantly lower than that in normal group (P = 0.004, P <0.01). In AA group, the expression of Oatp2a1 in normal + AA group was significantly lower than that in normal group (P = 0.032, P <0.05). Conclusions: Oatp2a1 may be one of the material bases for the body to participate in wet turbid transport. The changes of Oatp2a1 expression in the small intestine and liver of spleen deficiency rats suggest that small intestine, liver and other organs may play a role in the wet-turbid transport of spleen deficiency Important role, spleen transport wet turbid function, may include not only the gastrointestinal tract function, but also may include part of the liver function, worthy of further study.
