目的:检测人食管鳞状细胞癌组织中Cyr61、WISP3 mRNA及蛋白的表达。方法:采用实时荧光定量PCR和免疫组织化学染色法对186例食管鳞状细胞癌和癌旁正常组织中Cyr61、WISP3表达水平进行测定。采用Kaplan-Meier进行单因素生存分析。结果:食管鳞状细胞癌组织中Cyr61、WISP3 mRNA和蛋白的表达较正常组织上调(P均<0.01)。食管鳞状细胞癌组织中WISP3和Cyr61 mRNA的表达与淋巴结转移、浸润深度、肿瘤体积、肿瘤分期有关(P均<0.01),WISP3和Cyr61蛋白表达与吸烟、淋巴结转移、家族史、浸润深度和肿瘤分期有关(P均<0.05)。单因素生存分析表明,WISP3、Cyr61 mRNA和蛋白的表达、性别、吸烟、家族史、浸润深度、肿瘤体积、肿瘤分期与患者的生存率相关(P均<0.05)。结论:Cyr61、WISP3在食管鳞状细胞癌发展过程中起重要作用;其在食管鳞状细胞癌病情进展的监测、转移潜能及预后的估计等方面具有应用价值。
Objective: To detect the expression of Cyr61 and WISP3 mRNA and protein in human esophageal squamous cell carcinoma. Methods: The expression of Cyr61 and WISP3 in 186 cases of esophageal squamous cell carcinoma and adjacent normal tissues were detected by real-time fluorescence quantitative PCR and immunohistochemical staining. One-way survival analysis using Kaplan-Meier. Results: The expressions of Cyr61 and WISP3 mRNA and protein in esophageal squamous cell carcinoma tissues were up-regulated compared with normal tissues (all P <0.01). The expressions of WISP3 and Cyr61 mRNA in esophageal squamous cell carcinoma were correlated with lymph node metastasis, depth of invasion, tumor volume and tumor stage (all P <0.01). The expression of WISP3 and Cyr61 protein correlated with smoking, lymph node metastasis, family history, depth of invasion and Tumor stage (P <0.05). Univariate survival analysis showed that the expression of WISP3, Cyr61 mRNA and protein, gender, smoking, family history, depth of invasion, tumor volume and tumor stage correlated with the survival rate of patients (all P <0.05). Conclusion: Cyr61 and WISP3 play an important role in the development of esophageal squamous cell carcinoma. They are valuable in monitoring the progression of esophageal squamous cell carcinoma, metastatic potential and prognosis.
