目的:建立桑椹颗粒中绿原酸的含量测定方法。方法:采用薄层鉴别法(TLC)对制剂中绿原酸进行定性鉴别;以HPLC法测定桑椹颗粒中绿原酸的含量,采用Kromasil C_(18)色谱柱(250 mm×4.6 mm,5μm);使用乙腈-3m L·L~(-1)磷酸水溶液(15∶85)为流动相进行梯度洗脱;流速为0.8 m L·min~(-1);以327 nm为检测波长;柱温为室温。结果:绿原酸的薄层鉴别具有良好的专属性;绿原酸的回归方程:Y=42256X-9219.6,r=0.9994,线性范围为1.300~312.000μg·m L~(-1);平均回收率为96.62%,RSD%值为1.05%。结论:本文建立了桑椹颗粒中绿原酸的HPLC含量测定方法,结果准确,重复性好,可作为桑椹颗粒的质量评价方法。
Objective: To establish a method for determination of chlorogenic acid in mulberry leaves. METHODS: Chlorogenic acid was identified by TLC. The content of chlorogenic acid in mulberry leaves was determined by HPLC. The content of chlorogenic acid was determined by Kromasil C 18 column (250 mm × 4.6 mm, 5 μm) The mobile phase was eluted with acetonitrile-3m L · L -1 phosphoric acid (15:85). The flow rate was 0.8 m L · min -1. The detection wavelength was 327 nm. The column temperature For room temperature. Results: The TLC of chlorogenic acid had good specificity. The regression equation of chlorogenic acid was Y = 42256X-9219.6, r = 0.9994, the linear range was 1.300 ~ 312.000μg · m L -1; The rate was 96.62% and the RSD% was 1.05%. Conclusion: The HPLC method for the determination of chlorogenic acid in mulberry leaves is established in this paper. The method is accurate and reproducible. It can be used as a quality evaluation method for mulberry granules.
