目的:建立高效液相色谱法测定比卡硫醚中邻氟异构体含量。方法:采用Shim-packVP-ODS(250mm×4.6mm,5μm)色谱柱,四氢呋喃-水(45∶55)为流动相,272nm波长处检测。结果:比卡硫醚及其异构体达到基线分离,分离度为1.7,邻氟异构体在1~20mg·mL-1范围内线性关系良好(r=0.9999),平均回收率为99.5%,RSD为0.9(n=9),定量限2ng。结论:本方法能有效分离比卡硫醚及其异构体,方法简便,准确度好,精密度高。
Objective: To establish a HPLC method for the determination of o-fluoroisomers in pika sulfamethoxazole. Methods: Shim-pack VP-ODS (250mm × 4.6mm, 5μm) column and tetrahydrofuran-water (45:55) were used as mobile phase and detected at 272nm. RESULTS: Bika thioether and its isomers were baseline separated with a resolution of 1.7. The linear relationship between o-fluoroisomers was in the range of 1 ~ 20 mg · mL-1 (r = 0.9999) and the average recovery was 99.5% , RSD was 0.9 (n = 9), and the limit of quantification was 2 ng. Conclusion: The method can effectively separate pika thioether and its isomers. The method is simple, accurate and precise.
