本文旨在研究自体骨髓移植中多量骨髓的冷冻保存问题。(1)比较了正常骨髓粒系定向造血干细胞(CFU_(GM))活力的三种测试方法。在体内扩散盒培养方法中,用环磷酰胺0.3mg/g代替照射处理小鼠。(2)11例正常骨髓标本冻存于液氮并定期复温观察有核细胞回收率、锥蓝拒染率及CFU_(GM)回收率。当骨髓标本长期冻存达50周时,上述指标未见明显下降。(3)用二步离心法浓集多量骨髓的有核细胞。有核细胞可回收82.5±2.6%,所得有核细胞体积仅为原骨髓体积的三分之一左右。(4)在上述实验研究基础上,本文冻存了一例宫颈癌患者的自体骨髓。一个月后回输给患者的有核细胞总数为0.53×10~(10),CFU_(GM)总数为0.27×10~6。输注后患者的情况良好。
This article aims to investigate the cryopreservation of large amounts of bone marrow in autologous bone marrow transplantation. (1) Three test methods comparing the activity of normal bone marrow-derived granulocyte-directed hematopoietic stem cells (CFU_ (GM)) were compared. In the in vivo diffusion cassette culture method, mice were treated with cyclophosphamide 0.3 mg / g instead of irradiation. (2) Eleven patients with normal bone marrow were frozen in liquid nitrogen and observed for recovery of nucleated cells, blue-chip rejection rate and recovery of CFU_ (GM). When bone marrow samples were cryopreserved for up to 50 weeks, there was no significant decrease in the above indexes. (3) using two-step centrifugation to collect large amounts of bone marrow nucleated cells. Nucleated cells can be recovered 82.5 ± 2.6%, the volume of nucleated cells obtained is only about one-third of the original bone marrow volume. (4) On the basis of the above experimental study, a frozen cervical autologous bone marrow of patients with cervical cancer. The total number of nucleated cells returned to the patients after one month was 0.53 × 10 ~ (10), and the total number of CFU_ (GM) was 0.27 × 10 ~ 6. The condition of the patient after the infusion is good.
