本文报导棉酚对大鼠睾丸初级精母细胞DNA 含量的影响和体外抑制DNA合成试验的结果。大鼠服棉酚(30毫克/公斤/日)4及6周后,初级精母细胞核中DNA 的含量较对照组明显减少。其含量分布直方图主峰左移,不再呈典型的正态分布。体外抑制试验表明,当反应液中棉酚浓度为6微克分子时,DNA 合成受抑制率为12%,20微克分子时为20%,60微克分子时抑制率达最高峰值(23%)。根据上述结果,对棉酚干扰生精过程中DNA 代谢及其与抗生育作用的可能关系进行了分析和讨论。
This article reports the effect of gossypol on the DNA content of testicular primary spermatocytes in rats and the results of in vitro DNA synthesis assays. At 4 and 6 weeks after gossypol inhalation (30 mg / kg / day), the DNA content of primary spermatocyte nucleus was significantly reduced compared with the control group. The content distribution histogram shift to the left of the main peak, no longer a typical normal distribution. In vitro inhibition assay showed that when the concentration of gossypol in the reaction solution was 6 micromolar, the inhibition rate of DNA synthesis was 12%, 20% at 20 micromolar and the highest peak (23%) at 60 micromolar. Based on the above results, the possible relationship between gossypol interference with DNA metabolism and its anti-fertility effects during spermatogenesis was analyzed and discussed.
