目的 了解上海地区汉族人群 MICA基因第 5外显子微卫星多态性分布 ,以及 MICA基因与其紧密连锁的 HL A- B基因位点的关系。方法 用 PCR-异源双链分析法 ,对 175名正常无关个体的MICA基因第 5外显子微卫星多态性的分布进行研究。结果 (1)上海地区汉族人群 MICA基因第 5外显子存在 5种等位基因 ,其中 MICA* A5最为常见 (39.14% ) ,其次为 * A5 .1(2 2 .2 9% ) ;(2 ) MICA等位基因与 HL A- B位点之间存在一定的连锁不平衡 ,如 :MICA* A5与 HL A- B15 ,MICA* A4与 HL A- B2 7等。结论 MICA基因第 5外显子多态性分布虽没有 HL A经典 类、 类基因那样明显的人种和地区差异 ,但各群体之间仍然有差异存在
Objective To understand the distribution of microsatellite locus 5 exon of MICA gene and the relationship between MICA gene and its closely linked HLA-B locus in Shanghai Han population. Methods The distribution of exon 5 microsatellite polymorphism of MICA gene in 175 unrelated individuals was studied by PCR-heteroduplex analysis. Results (1) There were five alleles in exon 5 of MICA gene in Shanghai Han population, of which MICA * A5 was the most common (39.14%), followed by A5.1 (22.29%); MICA * A5 and HL A-B15, MICA * A4 and HL A-B2 7, etc. There are some linkage disequilibrium between MICA allele and HL A-B locus. Conclusion Although the distribution of exon 5 of MICA gene is not as obvious as ethnic and regional differences in HL A class and class genes, there are still some differences between the groups
