目的:探讨甘草抗病毒有效部位(GC3-1-4)体外抑制呼吸道合胞病毒(RSV)的作用。方法:采用煎煮法、乙醇沉淀法和柱层析法分离提取GC3-1-4。以病毒唑为阳性对照药物,采用细胞培养技术观察GC3-1-4抑制RSV作用。结果:GC3-1-4的TC50为0.45mg/ml,抑制RSV的EC50为41.32μg/ml,TI为10.89。GC3-1-4对RSV的抑制作用存在量效关系;感染RSV后0h、2h、4h、6h、8h加药,GC3-1-4在RSV的活性有抑制作用。化学实验证实GC3-1-4不含甘草酸或多糖类物质。结论:GC3-1-4在Hela细胞中对RSV有明显的抑制作用。
Objective: To investigate the anti-respiratory syncytial virus (RSV) effect of Glycyrrhiza uralensis active ingredient (GC3-1-4) in vitro. Methods: GC3-1-4 was isolated and extracted by decoction, ethanol precipitation and column chromatography. With ribavirin as a positive control drug, cell culture techniques were used to observe the effect of GC3-1-4 on RSV inhibition. RESULTS: The TC50 of GC3-1-4 was 0.45 mg/ml, the EC50 of RSV was 41.32 μg/ml, TI was 10.89. There was a dose-effect relationship between the inhibitory effect of GC3-1-4 on RSV. After RSV was given 0h, 2h, 4h, 6h, and 8h, GC3-1-4 inhibited the activity of RSV. Chemical experiments confirmed that GC3-1-4 contained no glycyrrhizic acid or polysaccharides. Conclusion: GC3-1-4 has obvious inhibitory effect on RSV in Hela cells.
