目的探讨空肠弯曲菌(Campylobacter jejuni,CJ)pcDNA3.1(-)-peb1A疫苗免疫昆明种小鼠后T细胞活化增殖状态。方法重组质粒CJ pcDNA3.1(-)-peb1A免疫昆明种小鼠后,取其脾淋巴细胞,应用ELISA法检测其膜表面分子CD3、CD4、CD8、CD25和CD28。结果检测结果显示:CD3检测,佐剂DNA组高于裸DNA组,在第10天有显著性差异(P<0.05);CD8检测,佐剂DNA组高于裸DNA组,在第10、20天均有显著性差异(P<0.05),裸DNA组也高于两对照组,但仅在第20天有显著性差异(P<0.05);CD25检测,裸DNA组高于NS组,但仅在第10天有显著性差异(P<0.05),佐剂DNA组高于裸DNA组,也仅在第20天有显著性差异(P<0.05);CD4及CD28检测,佐剂DNA组高于裸DNA组,在第10、20天均有显著性差异(P<0.05);在第10、20天各脾淋巴细胞膜表面分子检测,佐剂DNA组高于两对照组,有显著性差异(P<0.05)。结论 CJ pcDNA3.1(-)-peb1A疫苗能够有效地诱导T细胞活化增殖。
Objective To investigate the T lymphocyte activation and proliferation of Kunming mice injected with Campylobacter jejuni (CJ) pcDNA3.1 (-) - peb1A vaccine. Methods Immunized Kunming mice with recombinant plasmid CJ pcDNA3.1 (-) - peb1A and their splenic lymphocytes were harvested. The surface molecules CD3, CD4, CD8, CD25 and CD28 were detected by ELISA. Results The results of CD3 test and adjuvant DNA group were higher than those of naked DNA group at day 10 (P <0.05). CD8 and adjuvant DNA group were higher than those of naked DNA group, (P <0.05). There were also significant differences between the naked DNA group and the control group on the 20th day (P <0.05). The levels of CD25 in the naked DNA group were higher than those in the NS group Only on the 10th day (P <0.05), the DNA of the adjuvant group was higher than that of the naked DNA group, and there was a significant difference at the 20th day (P <0.05) (P <0.05). On the 10th and 20th days, the molecular detection of the spleen lymphocyte membrane surface was higher in the adjuvant DNA group than in the two control groups, with significant difference Difference (P <0.05). Conclusion CJ pcDNA3.1 (-) - peb1A vaccine can effectively induce T cell activation and proliferation.
