目的比较大理猪源旋毛虫(Trichinela spiralis)成虫与肌幼虫表面抗原(Surface antigen,SA)的蛋白组分差异,并进一步分析其免疫反应性,为分离和筛选出免疫原性和免疫反应性强的旋毛虫抗原成分提供依据。方法采用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE),对成虫和肌幼虫SA蛋白成分进行分析,并进一步采用蛋白印迹(Western-blot)对其免疫反应性分析比较。结果旋毛虫成虫SA显示4条主要蛋白染色条带,分子量分别为50kDa、48kDa、40kDa和34kDa,旋毛虫肌幼虫SA显示10条主要蛋白染色条带,分别为105kDa、99kDa、80kDa、63kDa、55kDa、48kDa、35kDa、28kDa、24kDa和21kDa。;Western-blot检测结果表明,旋毛虫成虫SA出现4条反应条带,其分子量分别为50kDa、48kDa、40kDa和34kDa,旋毛虫肌幼虫SA出现7条反应条带,分子量分别为105kDa、99kDa、80kDa、55kDa、48kDa、35kDa和28kDa,其中48kDa和35kDa显色明显。结论旋毛虫成虫SA与肌幼虫SA都含有分子量为48kDa的蛋白组分,且两种抗原组分均具有较强的免疫反应性。
Objective To compare the protein components of Trichinella spiralis adults and muscle larvae with surface antigen (SA), and to further analyze the immunoreactivity of Trichinella spiralis in order to isolate and screen out the immunogenicity and immunoreactivity Trichinella antigen components provide the basis. Methods The protein components of adult and muscle larvae were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), and their immunoreactivity was further analyzed by Western-blot. Results The adult Trichinella spiralis showed four major protein bands with the molecular weights of 50kDa, 48kDa, 40kDa and 34kDa, respectively. SA of Trichinella spiralis larvae showed 10 major protein bands with 105kDa, 99kDa, 80kDa, 63kDa and 55kDa , 48 kDa, 35 kDa, 28 kDa, 24 kDa and 21 kDa. ; Western-blot results showed that there were 4 reaction bands in the SA of Trichinella spiralis, the molecular weights were 50kDa, 48kDa, 40kDa and 34kDa, respectively. There were 7 reaction bands in SA of Trichinella spiralis larvae with molecular weights of 105kDa and 99kDa, 80kDa, 55kDa, 48kDa, 35kDa and 28kDa, of which 48kDa and 35kDa color obvious. Conclusion SA and muscle larvae SA of Trichinella spiralis contain a molecular weight of 48kDa protein components, and both antigen components have strong immunoreactivity.
