抗人β防御素3融合蛋白抗体的制备及其活性检测

目的:人抗菌肽β防御素3(HBD3)除具有杀灭多种病原菌的作用外,还参与机体免疫防御的多个过程,本实验的目的是进一步了解HBD3在人体的生物功能.方法:通过将编码HBD3成熟链的基因与载体pGEX-KG相连,表达出谷胱甘肽S转移酶与HBD3(GST-HBD3)的融合蛋白,制备HBD3的抗体.结果:通过Western blot检测及免疫组化方法证实HBD3抗体的制备成功,显示了HBD3蛋白在食道上皮细胞的表达部位.结论:为进一步了解HBD3在人体的功能及其在疾病状态下的改变奠定了坚实的基础.
