本文建立了快速、简便测定人血清中氨氯地平的HPLC法,分析柱为 ODS-5S( 150 × 4.0mm)不锈钢柱,流动相为甲醇:0.03M磷酸二氢钾缓冲液(9∶1)的混合液。流速为1.0ml/min,以硝苯地平为内标物,检测波长为366nm,蛋白沉淀剂选用15%高氨酸。氨氯地平测定的线性范围为4~12μg/ml,r=0.9992,最低检测限度为 4ng。日内平均相对回收率为 10.24 ± 5.40%,日间平均相对回收率为 99.87 ± 6.23%。本方法专一性好,放置24h对含量测定无影响。
In this paper, a rapid and simple HPLC method for the determination of amlodipine in human serum was established. The analytical column was ODS-5S (150 × 4.0 mm) stainless steel column. The mobile phase consisted of methanol: 0.03 M potassium dihydrogen phosphate buffer (9: 1). Flow rate of 1.0ml / min to nifedipine as the internal standard, the detection wavelength of 366nm, 15% selection of protein precipitator of high-acid. Amlodipine determination of the linear range of 4 ~ 12μg / ml, r = 0.9992, the minimum detection limit of 4ng.
