中药含药血浆和血清对DMEM培养液渗透压和PH值的影响

目的:观察不同来源的单味和复方中药延胡索、水蛭、朱砂、四物汤的含药血浆和含药血清对DMEM培养液的渗透压和PH值影响,探讨中药血浆药理方法体外反应体系构建。方法:通过在DMEM培养液中加入延胡索、水蛭、朱砂、四物汤的含药血浆与含药血清,按照细胞培养的常规条件进行培养,分别测定其培养第1、2、4、7天后培养液的冰点渗透压和PH值。结果:随着培养时间的延长,各组别培养液的渗透压和PH值逐渐增加。但与同期对照组比较,加入含药血清与含药血浆培养液的渗透压和PH值差异不具有显著性(P>0.05)。同期相同药物和剂量的含药血浆与含药血清培养液的渗透压和PH值之间差异同样不具有显著性(P>0.05)。结论:不同来源的单味和复方中药的含药血浆与含药血清对细胞培养液的渗透压和PH值影响基本一致,提示中药半体内实验中单味中药或复方的含药血浆和含药血清均不影响细胞培养体系的渗透压和PH值。 OBJECTIVE: To observe the effect of drug-containing plasma and drug-containing serums of Cynanchum, leech, vermilion, Siwu decoction from different sources on the osmotic pressure and pH value of DMEM culture medium and to explore the in vitro reaction system construction of traditional Chinese medicine pharmacology and pharmacology. Methods: The drug-containing plasma of Corydalis yanhusuo, leeches, cinnabar and Siwu decoction and the serum containing drug were added into DMEM culture medium and cultured according to the conventional conditions of cell culture. The culture was carried out on days 1, 2, 4 and 7 Fluid freezing point osmotic pressure and PH value. Results: With the prolongation of culture time, the osmotic pressure and pH value of culture medium in each group increased gradually. However, compared with the control group at the same period, there was no significant difference in osmotic pressure and pH between the drug-containing serum and drug-containing plasma (P> 0.05). At the same period, the difference between the osmotic pressure and pH value of drug-containing plasma and drug-containing serum medium of the same drug and dose was also not significant (P> 0.05). CONCLUSION: The effects of drug-containing plasma and drug-containing serum of single-sourced and compound Chinese herbs from different sources on the osmotic pressure and pH value of cell culture medium are basically the same, suggesting that single-drug or compound-containing drug-containing plasma and drug- Serum does not affect the cell culture system of osmotic pressure and PH value.
