目的探讨不同病理学级别人脑星形细胞瘤中Rho GTPases相关信号通路蛋白Wis-kott-Aldrich综合征蛋白质(WASP)与T淋巴瘤侵袭转移诱导因子1(Tiam1)所扮演的可能角色。方法应用免疫组织化学法检测60例不同病理级别人脑星形细胞瘤组织中WASP、Tiam1蛋白表达,评估其与星形细胞瘤的病理分级及临床特征的关系。结果 (1)Tiam1蛋白高级别组的阳性率和高表达率均高于瘤旁脑组织和低级别组(P<0.05),且Tiam1蛋白的阳性率和高表达率有从瘤旁脑组织→低级别星形细胞瘤→高级别星形细胞瘤递增的趋势,Tiam1表达的强弱在一定程度上反映了人脑星形细胞瘤的恶性程度。(2)Tiam1蛋白表达的阳性率和高表达率与年龄、性别、部位及肿瘤最大径无关(P>0.05)。(3)WASP蛋白为阴性表达。结论 WASP蛋白与星形细胞瘤发生和进展的相关性不明确;Tiam1蛋白在了解星形细胞瘤恶性进展方面有参考价值,且可能是有预后价值因子之一。
Objective To investigate the possible role of Rho GTPases-related signaling pathway protein Wis-kott-Aldrich syndrome protein (WASP) and T lymphoma invasion and metastasis-inducible factor 1 (Tiam1) in human pathologic grade human brain astrocytomas. Methods The expression of WASP and Tiam1 protein in 60 cases of human astrocytomas with different pathological grades were detected by immunohistochemistry. The relationship between WASP and Tiam1 protein expression was analyzed with the pathological grade and clinical features of astrocytomas. Results (1) The positive rate and high expression rate of Tiam1 protein in high-grade group were higher than those in the tumor-adjacent brain tissue and low-grade group (P <0.05), and the positive rate and high expression rate of Tiam1 protein from the paracancerous brain tissue → Low-grade astrocytoma → high-grade astrocytoma increasing trend, the intensity of Tiam1 expression to some extent, reflects the degree of malignancy of human brain astrocytoma. (2) The positive rate and high expression rate of Tiam1 protein had no relation with age, sex, location and tumor diameter (P> 0.05). (3) WASP protein was negative expression. Conclusion The relationship between WASP protein and the occurrence and progression of astrocytomas is not clear. Tiam1 protein is valuable in understanding the malignant progression of astrocytomas and may be one of the prognostic factors.
