目的研究脑神康胶囊对糖尿病大鼠记忆能力及海马乏氧诱导因子(HIF-1α)表达的影响。方法将36只大鼠分为正常对照组、糖尿病组、中药治疗组,中药治疗组用脑神康胶囊水溶液灌胃,其余两组用等量生理盐水灌胃,每日1次,持续12周。在0、4、8、12周测血糖及进行Morris水迷宫实验,使用免疫组化及RT-PCR技术检测海马HIF-1α的表达。结果中药治疗组与糖尿病组大鼠HIF-1α表达量较正常对照组增高,中药治疗组第8、12周较糖尿病组HIF-1α表达量减少(P<0.01或P<0.05)。水迷宫潜伏期自第4周始中药治疗组较糖尿病组缩短(P<0.01或P<0.05),较正常对照组延长。结论脑神康胶囊能下调HIF-1α表达,改善记忆能力下降症状,减轻糖尿病脑功能损害。
Objective To study the effect of “Aoqikang Capsule” on the memory ability and hypoxia-inducible factor (HIF-1α) expression of hippocampus in diabetic rats. Methods Thirty-six rats were divided into normal control group, diabetic group, traditional Chinese medicine treatment group and traditional Chinese medicine treatment group by intragastric administration of Naosinkang capsule. The other two groups were given intragastrically with normal saline once a day for 12 weeks . Blood glucose was measured at 0, 4, 8 and 12 weeks and Morris water maze test was performed. The expression of HIF-1α in hippocampus was detected by immunohistochemistry and RT-PCR. Results The expression of HIF-1α in Chinese medicine group and diabetic group was higher than that in normal control group. The expression of HIF-1α in Chinese medicine group was lower than that in diabetic group at the 8th and 12th weeks (P <0.01 or P <0.05). The latency of water maze was shorter than that of the diabetic group (P <0.01 or P <0.05) from the 4th week onwards, which was longer than that of the normal control group. CONCLUSION Naoxinkang Capsule can down-regulate the expression of HIF-1α, improve the symptoms of memory loss and reduce the damage of diabetic brain function.
