目的:观察健脾燥湿经典方苍白二陈汤对代谢综合征(MS)大鼠主动脉ICAM1表达的影响,探讨其降低动脉粥样硬化风险的可能分子机制。方法:以高糖高脂高盐饲料长期喂饲建立MS大鼠模型。将21只MS大鼠随机分为模型组、西药组和中药组,分别予蒸馏水、洛伐他汀和苍白二陈汤灌胃。4周后,测血脂(TG、TC、LDLc、HDLc和ox-LDL),计算动脉粥样硬化指数(AI),免疫组化法检测主动脉ICAM1表达。结果:与模型组相比,中药组大鼠血清TG、TC、LDLc和ox-LDL水平,AI及主动脉ICAM1表达量(以平均光密度表示)均显著降低(P<0.05)。结论:苍白二陈汤可以调节代谢综合征模型大鼠的血脂,降低AI,抑制主动脉ICAM1的表达,具有抗动脉粥样硬化作用。
Objective: To observe the effect of Jianpi dampness Decoction of Fangfang Erchen Decoction on the expression of ICAM1 in the aorta of rats with metabolic syndrome (MS) and to explore its possible molecular mechanism of reducing the risk of atherosclerosis. Methods: MS rat model was established by long-term feeding with high-sugar, high-fat and high-salt diet. Twenty-one MS rats were randomly divided into model group, western medicine group and traditional Chinese medicine group, respectively, with distilled water, lovastatin and pale Erchen decoction. After 4 weeks, the blood lipids (TG, TC, LDLc, HDLc and ox-LDL) were measured to calculate the atherosclerosis index (AI). The expression of ICAM1 in the aorta was detected by immunohistochemistry. Results: Compared with the model group, the levels of TG, TC, LDLc and ox-LDL, the expression of ICAM1 in AI and aorta (mean optical density) of Chinese medicine group were significantly decreased (P <0.05). Conclusion: Wanbachen decoction can regulate the blood lipid of rats with metabolic syndrome, reduce the AI, inhibit the expression of ICAM1 in the aorta, and has the anti-atherosclerotic effect.
