目的为深入探讨红景天苷对镉中毒的保护作用及其机制,研究红景天苷在镉致BRL 3A细胞线粒体凋亡途径中的作用。方法 10μmol/L醋酸镉和50μmol/L红景天苷(Salidroside,SAL)单独或联合作用肝细胞(BRL 3A)12 h,采用实时分析技术(RTCA)测定细胞指数(CI),Hoechst 33258染核、流式细胞术检测细胞凋亡,western blot法检测Bax、Bcl-2、caspase-3蛋白表达和细胞色素C(cytochrome c,Cyt C)的释放,研究红景天苷对肝细胞(BRL 3A)的保护作用。结果红景天苷改善镉引起的CI值下降(P<0.05),降低镉引起的细胞凋亡率的升高,抑制镉引起的Bax/Bcl-2比值和cleaved caspase-3/procaspase-3比值升高(P<0.05或P<0.01),抑制线粒体Cyt C的释放(P<0.01)。结论红景天苷抑制镉诱导的BRL 3A细胞凋亡,可能与线粒体凋亡途径有关。
Objective To explore the protective effect of salidroside on cadmium poisoning and its mechanism, and to investigate the role of salidroside in mitochondrial apoptotic pathway of BRL 3A cells induced by cadmium. Conclusion Salidroside can inhibit the apoptosis of BRL 3A cells induced by cadmium, which may be related to the mitochondrial apoptosis pathway.
