目的分离和电镜鉴定EG.7小鼠胸腺肿瘤细胞分泌的exosom es,并观察其抗肿瘤作用。方法常规分级离心分离和密度梯度离心纯化得到exosom es,并对exosom es沉淀行透射电镜鉴定。应用分离纯化的exosom es免疫小鼠然后荷瘤,观察肿瘤的生长状况。结果EG.7细胞来源的exosom es大小40~100nm,具有膜结构。exosom es免疫能够显著抑制肿瘤生长和延长荷瘤小鼠的存活期。结论从肿瘤细胞培养上清中分离纯化的exosom es具有显著的抗肿瘤作用。
Objective To isolate and identify the exosom es secreted by the thymus tumor cells of EG.7 mice by electron microscopy and observe its antitumor effect. Methods Conventional fractionated centrifugation and density gradient centrifugation were used to obtain exosom es. The exosom es precipitates were identified by transmission electron microscopy. Immunize mice with isolated and purified exosom es and then tumor-bearing mice to observe the growth of the tumor. Results The exosom es derived from EG.7 cells were 40-100 nm in size and had membrane structure. Exosom es immunization can significantly inhibit tumor growth and prolong the survival of tumor-bearing mice. Conclusion The purified exosom es from tumor cell culture supernatant has a significant anti-tumor effect.
