目的:探讨鼻咽癌细胞增殖活性指标有丝分裂指数(mitotic index,MI)、增殖细胞核抗原(proliferating cell nuclear anti—gen,PCNA)、p53蛋白、DNA含量和癌细胞核形态参数与放射敏感性的关系。方法:收集51例放疗前鼻咽癌组织标本,HE染色作病理组织学诊断和MI计数;免疫组化法检测PCNA和p53蛋白表达情况;Feulgen染色和图像分析法检测鼻咽癌DNA含量和细胞核的形态定量参数。结果:51例鼻咽癌组织中,完全消退组MI高于残留组(P<0.05);PCNA和p53蛋白表达阳性音分别有50例(98.0%)和46例(90.2%),高表达者分别有16例(31.4%)和22例(43.1%);PCNA和p53蛋白的阳性表达水平越高,则鼻咽癌对放射治疗越敏感,放疗后鼻咽局部癌肿完全消退率越高(P<0.05);PCNA和p53蛋白同时高表达者比单一指标高表达者,对其放射敏感性更高(P<0.05);鼻咽癌完全消退组DI、DNA异倍体细胞所占百分率、细胞核形态参数面积、周长和直径均比残留组高。结论:MI、PCNA、p53蛋白、DNA含量和癌细胞核形态参数是反映鼻咽癌放射敏感性和预测其近期疗效的重要指标。
Objective: To investigate the relationship between mitochondrial index (MI), proliferating cell nuclear anti-gen (PCNA), p53 protein, DNA content and nuclear morphological parameters and radiosensitivity of nasopharyngeal carcinoma cells. Methods: 51 cases of nasopharyngeal carcinoma before radiotherapy were collected for pathological diagnosis and MI count by HE staining. The expressions of PCNA and p53 protein were detected by immunohistochemistry. The DNA content and nucleus were detected by Feulgen staining and image analysis The morphological parameters. Results: In 51 cases of nasopharyngeal carcinoma, the MI of complete remission group was higher than that of the residual group (P <0.05). The positive expressions of PCNA and p53 were 50 (98.0%) and 46 %), Respectively. There were 16 cases (31.4%) and 22 cases (43.1%) with high expression respectively. The higher the expression of PCNA and p53 protein was, the more sensitive nasopharyngeal carcinoma was to radiation therapy. The complete remission rate of pharyngeal local cancer was higher (P <0.05); PCNA and p53 protein overexpression were higher than those with single overexpression (P <0.05); nasopharyngeal carcinoma Completely dissipated group DI, the percentage of DNA aneuploid cells, nuclear morphology parameter area, circumference and diameter were higher than the residual group. Conclusion: MI, PCNA, p53 protein, DNA content and nuclear morphological parameters of cancer cells are important indexes to reflect the radiosensitivity of nasopharyngeal carcinoma and predict its short-term curative effect.
