目的:研究猪来源抗胸腺细胞球蛋白(ATG)对食蟹猴免疫功能的影响。方法 ATG倍比稀释与食蟹猴淋巴细胞孵育,流式检测细胞毒;ATG单纯给药组分为低剂量组(250m g/日)及高剂量组(750m g/日),分别连续给药5日,并于定期检测血常规以及用流式细胞仪检测C D3/C D20、C D4/C D8细胞的比例。结果 ATG对食蟹猴淋巴细胞具有很强的杀伤作用,滴度为1:2时杀伤率为84.95%,直至滴度为1:512时才降至10%以下;给药期间体内单纯给药高剂量组与低剂量组对总淋巴细胞清除率无明显差别,但各组对CD8+淋巴细胞杀伤作用均强于CD4+淋巴细胞(P<0.01)。结论 ATG可以杀伤食蟹猴淋巴细胞,其中对CD8+淋巴细胞效果更显著。“,”Objective To investigate the influence of ATG on cynomolgus immune function. Methods Cynomolgus lymphocytes were incubated with various concentrations of ATG to detect cytotoxicity by flow cytometry; In ATG intravenous infusion experiment, cynomolgus are divided into two groups: low dosage group (250mg/day) or high dosage group (750mg/day) and recived intravenous infusion for 5 days. Blood samples were col ected for blood routine analysis and CD3, CD20, CD8 and CD4 percentage by flow cytometry. Results ATG has strong cytotoxicity effect against cynomolgus lymphocyte. When ATG titer was 1:2, the lysis rate was 84.95%, and the lysis rate dropped below 10% when the titer reached 1:512. There is no difference in in vivo ATG intravenous infusion experiment between High dosage group and low dosage group in general depletion rate during administration period. The CD8 positive cells depletion rate is stronger than the CD4 positive cells depletion rate. Conclusion ATG has a cytotoxity effect against cynomolgus lymphocytes.
