为深入了解血液常温保存中红细胞的损伤性变化,采用电子自旋共振(ESR)技术对常温(35℃)保存的人血样品,进行了血红蛋白氧化变性研究。结果显示:35℃保存的全血在保存36小时左右出现高自旋的高铁血红蛋白吸收峰(g=6),且随保存时间的增加而增大。在4℃保存的全血,连续60小时的保存实验均未发现有任何ESR波谱吸收峰出现。35℃保存人血红细胞中,超氧化物歧化酶的活性随保存时间的延长而逐步降低。血红蛋白的氧化变化,可能是由于红细胞内抗氧化系统在血液保存期间抗氧化效力逐步降低造成的。
In order to further understand the damage changes of erythrocytes during storage at room temperature, the hemoglobin oxidative degeneration of human blood samples stored at room temperature (35 ℃) was studied by electron spin resonance (ESR). The results showed that the high spinosylated hemoglobin peak (g = 6) appeared at 35 ℃ in whole blood for 36 hours and increased with the increase of storage time. Whole blood preserved at 4 ℃, 60 consecutive days of preservation experiments did not find any ESR spectrum absorption peak appears. 35 ℃ stored in human red blood cells, the activity of superoxide dismutase gradually decreased with the preservation of time. Oxidative changes of hemoglobin may be due to the gradual decrease of anti-oxidant efficacy of the intra-erythrocyte anti-oxidant system during blood preservation.
