目的探讨益气活血中药复方对慢性阻塞性肺疾病(COPD)合并肺动脉高压(PH)大鼠血清缺氧诱导因子-1α(HIF-1α)、促红细胞生成素(EPO)的影响。方法将65只wistar雄性大鼠随机分为空白对照组、模型组、中药组、西药组。模型组、中药组、西药组大鼠在第1、14天时气管内滴注0.2ml脂多糖(200μg/μl),之后2~13,15~70天大鼠于自制动物熏吸箱中被动吸烟,予香烟烟熏30min/次(每次15支,第1、14天除外),2次/天,每周6天,连续10周。于模型评价成功后进行灌胃2周,结束后取血及肺组织,检测HIF-1α、EPO。结果与空白对照组比较,模型组大鼠血清中HIF-1α、EPO水平明显升高,具有显著差异性(P<0.05)。与模型组比较,中西药各治疗组大鼠血清中HIF-1α、EPO均有不同程度下降(P<0.05)。结论中药复方可能是通过抑制HIF-1α、EPO的表达而起到对COPD合并PH大鼠的防治作用。
Objective To investigate the effects of Yiqi Huoxue Compound on serum levels of hypoxia inducible factor-1α (HIF-1α), erythropoietin (EPO) in chronic obstructive pulmonary disease (COPD) and pulmonary hypertension (PH) rats. Methods Sixty-five male wistar rats were randomly divided into blank control group, model group, TCM group and western medicine group. Rats in the model group, the traditional Chinese medicine group and the western medicine group were intratracheally instilled 0.2 ml lipopolysaccharide (200 μg / μl) on day 1 and day 14, and passive smoking was carried out in homemade animal smoked boxes after 2 to 13 and 15 to 70 days , Smoking cigarettes 30min / time (each 15, except the first and 14th days), 2 times / day, 6 days a week for 10 weeks. After successful model evaluation, gavage was performed for 2 weeks, and blood and lung tissues were taken after the model was completed. HIF-1α and EPO were detected. Results Compared with the blank control group, the serum levels of HIF-1α and EPO in the model group were significantly increased (P <0.05). Compared with the model group, the levels of HIF-1α and EPO in the serum of rats in the Chinese and western medicine groups decreased to different extents (P <0.05). Conclusion Traditional Chinese recipe may play a preventive and therapeutic role in COPD complicated with PH by inhibiting the expression of HIF-1α and EPO.
