目的探讨不同比活度的~(131)I标记大肠癌单抗片段对大肠癌细胞结合率的影响。方法应用37MBq的~(131)I分别标记1000、200、100、50、25μg的单抗片段,测定标记率>95%后与2×10~6的大肠癌细胞反应30min,离心、洗涤2次后测量结合率;对照组设骨髓瘤细胞,观察非特异性结合率;选取结合率最高组,增加细胞数,观察结合率变化。结果 1000、200、100、50、25μg的~(131)I大肠癌单抗片段的放射性比活度分别为37、185、370、740、1480kBq/μg;与2×10~6大肠癌细胞的结合率分别为10.1%、14.7%、35.1%、32.7%、22.6%;非特异性结合率为0.8%;增加细胞数可以进一步增加结合率。结论比活度为370~740 kBq/μg的碘标记大肠癌单抗片段对大肠癌细胞有较高的结合率。
Objective To investigate the effect of different specific activity of ~ (131) I labeled colorectal cancer monoclonal antibody on colorectal cancer cell binding rate. Methods ~ (131) I 37MBq were labeled with 1000, 200, 100, 50, 25μg of monoclonal antibody. After the labeling rate was> 95%, the cells were reacted with 2 × 10 ~ 6 colorectal cancer cells for 30min, The binding rate was measured. The control group was set up with myeloma cells to observe the nonspecific binding rate. The highest binding rate was selected to increase the number of cells and the binding rate was observed. Results The radioactive specific activity of ~ (131) I colorectal cancer cells in the range of 1000, 200, 100, 50 and 25μg were 37, 185, 370, 740 and 1480kBq / The binding rates were 10.1%, 14.7%, 35.1%, 32.7% and 22.6%, respectively. The nonspecific binding rate was 0.8%. Increasing the cell number could further increase the binding rate. Conclusion The specific activity of 370 ~ 740 kBq / μg of iodine-labeled colorectal cancer monoclonal antibody fragment on colorectal cancer cells with a higher binding rate.
