目的:建立一种能反映人类慢性阻塞性肺病(COPD)自然发病过程的动物模型。方法:通过被动吸烟和多次气道内注入脂多糖的复合方法制作COPD大鼠模型,观察大鼠一般状态如体重、精神状态、呼吸、毛发、饮食、气道中痰液的分泌量等,并取肺组织行病理切片、免疫组化检查。结果:模型组大鼠体重未见明显下降,咳嗽、喘息症状不明显。病理切片结果提示:模型组气道破坏、炎症浸润不明显,空白组与模型组IL-8、IL-10、TNF-α免疫组化数据相比无明显差异性。结论:在屏障环境下,作者通过气道内滴入脂多糖和被动吸烟复合刺激法未能成功建立COPD大鼠模型。
Objective: To establish an animal model that reflects the natural history of human chronic obstructive pulmonary disease (COPD). Methods: COPD rat model was established by passive smoking and repeated intratracheal instillation of lipopolysaccharide, and the general state of rats such as body weight, mental status, respiration, hair, diet, airway sputum secretion and so on Lung tissue biopsy, immunohistochemistry. Results: The body weight of rats in the model group showed no significant decline, cough, wheezing symptoms were not obvious. The results of pathological examination showed that there was no significant difference in airway destruction and inflammatory infiltration in the model group compared with the immunohistochemical data of IL-8, IL-10 and TNF-α in the blank group and the model group. CONCLUSION: In the barrier environment, the authors failed to establish a COPD rat model successfully by lipopolysaccharide infusion in the airway and passive smoking compound stimulation method.
