试验以H99、综31和郑58三个玉米自交系的成熟胚为外植体,利用正交试验设计L9(34),对影响愈伤组织诱导的基因型、培养基、2,4-D浓度和Ag NO3浓度4个因素进行了研究。结果表明:各因素对初级愈伤诱导率影响大小为基因型>培养基>2,4-D浓度>Ag NO3浓度;对胚性愈伤诱导率影响大小为基因型>Ag NO3浓度>2,4-D浓度>培养基。诱导中最优组合为综31+10 mg/L Ag NO_3+MN+2 mg/L 2,4-D,继代中最优组合为综31+10 mg/L Ag NO3+MN+1 mg/L 2,4-D,其初级愈伤诱导率为87.41%,胚性愈伤诱导率为29.74%。
In this study, the mature embryos of three maize inbred lines H99, He 31 and Zheng 58 were used as explants. The effects of L9 (34) D concentration and AgNO3 concentration four factors were studied. The results showed that the effect of each factor on primary callus induction rate was genotype> medium> 2,4-D concentration> AgNO3 concentration; the effect on embryogenic callus induction rate was genotype> AgNO3 concentration> 2, 4-D concentration> medium. The best combination of induction was 31 + 10 mg / L Ag NO 3 + MN + 2 mg / L 2,4-D, and the best combination in the subsequent generations was 31 + 10 mg / L Ag NO 3 + MN + 1 mg / L 2,4-D, the primary callus induction rate was 87.41%, embryogenic callus induction rate was 29.74%.
