目的:研究宁心红杞胶囊对去势雌性大鼠的雌激素样作用机制。方法:采用去势雌性大鼠模型(OVX),以雌二醇(17β-estradiol,E2)作为阳性对照,给予宁心红杞胶囊2周后,测定其子宫系数、血清E2含量;以免疫组化法观察子宫上血管内皮生长因子(vascular endothelial growth factor,VEGF)、水通道蛋白(aquaporin,AQP)、雌激素受体(estrogen receptor,ER)、孕激素受体(progesterone receptor,PR)和原癌基因c-jun,c-fos的表达;以酵母双杂交法检测宁心红杞胶囊中是否含有与ERα或ERβ结合的成分。结果:宁心红杞胶囊与E2均能显著增加OVX大鼠子宫系数,提高血清E2水平,上调子宫上VEGF,AQP2和AQP5的表达;但与E2不同,未见宁心红杞胶囊有促进OVX大鼠子宫上ERα,PR,c-jun,c-fos表达的作用。酵母双杂交实验未检测到宁心红杞胶囊中含有与ERα或ERβ结合的成分。结论:宁心红杞胶囊具有雌激素样作用,能治疗围绝经期综合征,但作用机制不同于雌激素类物质,较少发生雌激素介导的子宫内膜癌的风险。
Objective: To study the estrogen-like mechanism of Ningxinhongqi capsule in ovariectomized female rats. Methods: The ovariectomized female rat model (OVX) was used. The estradiol (E2) was used as the positive control. After 2 weeks of treatment, the uterus coefficient and serum E2 level were determined. The levels of vascular endothelial growth factor (VEGF), aquaporin (AQP), estrogen receptor (ER), progesterone receptor (PR) The expression of oncogene c-jun, c-fos was detected by yeast two-hybrid method. The composition of Ningxinhongqi capsule containing ERα or ERβ was detected. Results: Ningxinhongqi Capsule and E2 can significantly increase the uterus coefficient of OVX rats, raise the level of serum E2 and upregulate the expression of VEGF, AQP2 and AQP5 in uterus. However, unlike Ningxiang Red Qi Capsule, The role of ERα, PR, c-jun, c-fos expression in rat uterus. Yeast two-hybrid experiments did not detect Ningxin red Qi capsule containing ERα or ERβ binding components. Conclusion: Ningxinhongqi capsule has estrogen-like effect and can treat perimenopausal syndrome. However, its mechanism of action is different from that of estrogen and less risk of estrogen-mediated endometrial cancer.
