目的 明确我国分离的XJ 90 2 6 0和XJ 910 0 6病毒的分类地位、种系发生和遗传型。方法 特异引物逆转录 聚合酶链反应 (RT PCR)扩增两株病毒的NSP4、E1基因区和 3′端非编码区 ,测序 ,进行核苷酸序列同源性比较和 3′端非编码区核苷酸序列分析 ,并结合同属其他病毒这些基因区的核苷酸序列进行种系进化分析。结果 两株病毒的核苷酸序列同源性为 10 0 % ,与西方马脑炎病毒的核种酸序列同源性最高 ,具有西方马脑炎病毒 3′端非编码区结构特征 ,其NSP4基因区与东方马脑炎病毒同源 ,E1基因区与辛德毕斯病毒同源 ,两株病毒均位于西方马脑炎病毒B组 ,与西方马脑炎病毒俄罗斯分离株进化关系最近。结论 我国分离的XJ 90 2 6 0和 910 0 6病毒属于西方马脑炎病毒同一遗传型 ,均为重组甲病毒。
Objective To clarify the taxonomic status, phylogenetic and genetic patterns of XJ 90 2 60 and XJ 910 0 6 viruses isolated in China. METHODS: NSP4, E1 and 3 ’non-coding regions of the two viruses were amplified by reverse transcription-polymerase chain reaction (RT PCR) and sequenced. The nucleotide sequence homology and 3’ non-coding region Nucleotide sequence analysis and phylogenetic analysis based on the nucleotide sequences of these gene regions belonging to other viruses. Results The nucleotide sequence homology of the two viruses was 100%. The two strains shared the highest homology with the western equine encephalitis virus and the non - coding region of the western equine encephalitis virus. The NSP4 The gene region is homologous to the equine Eastern equine encephalitis virus. The E1 gene region is homologous to the Sindbis virus. Both viruses are located in the western equine encephalitis virus group B and have the closest evolutionary relationship with the western equine encephalitis virus strain Russia. Conclusion The XJ 90 2 60 and 910 0 6 viruses isolated in China belong to the same genetic type of western equine encephalitis virus and all are recombinant alphavirus.
