目的:观察拓扑替康(TPT)对宫颈癌Hele细胞增殖与凋亡的影响。方法:在相同放疗条件下,采用MTT法检测不同浓度TPT对HeLa细胞增殖的抑制作用;采用流式细胞仪检测细胞周期,TUNEL法检测TPT对HeLa细胞凋亡的影响。结果:随着TPT浓度升高(0.05 mg/L、0.1 mg/L、0.2 mg/L、升至0.4 mg/L),HeLa细胞的增殖抑制率逐渐升高,细胞的凋亡率依次增加。结论:TPT能抑制宫颈癌HeLa细胞的增殖并促进其凋亡。
Objective: To observe the effect of topotecan (TPT) on the proliferation and apoptosis of cervical cancer Hele cells. Methods: MTT assay was used to detect the inhibitory effect of different concentrations of TPT on HeLa cell proliferation under the same radiotherapy conditions. The cell cycle was detected by flow cytometry. The apoptosis of HeLa cells was detected by TUNEL assay. Results: With the increase of TPT concentration (0.05 mg / L, 0.1 mg / L, 0.2 mg / L, up to 0.4 mg / L), the proliferation inhibition rate of HeLa cells gradually increased and the apoptosis rate of HeLa cells increased in turn. Conclusion: TPT can inhibit the proliferation and promote the apoptosis of cervical cancer HeLa cells.
