目的探讨隆凸性皮肤纤维肉瘤(DFSP)中CD34、血小板源性生长因子受体(PDGFR-β)的表达及基因倍体的意义。方法使用免疫组化结合图象分析法检测39例DFSP,30例皮肤纤维瘤(DF)皮损中CD34、PDGFR-β的表达情况。采用流式细胞技术分析样本细胞核基因倍体,计算其DI值。结果CD34、PDGFR-β蛋白在DFSP中表达明显高于在DF的表达,差异具有统汁学意义(P<0.05)。直线相关分析表明,CD34、PDGFR-β蛋白的表达之间无相关性(P>0.05)。39例DFSP中9例为异倍体(23%),其余为二倍体,异倍体均为近二倍体(DI=1.16±0.14);30例DF中无1例异倍体。相关性分析发现,异倍体组PDGFR-β蛋白的表达较二倍体组高(P<0.05)。结论 CD34、PDGFR-β与隆凸性皮肤纤维肉瘤的发生、发展密切相关。
Objective To investigate the expression of CD34, PDGFR-β and the significance of gene ploidy in Convex Dermal Fibrosarcoma (DFSP). Methods Immunohistochemistry and image analysis were used to detect the expression of CD34 and PDGFR-β in 39 cases of DFSP and 30 cases of dermal fibroma (DF). The flow cytometry was used to analyze the nuclear genome of the sample and calculate its DI value. Results The expressions of CD34 and PDGFR-β in DFSP were significantly higher than those in DF, with significant differences (P <0.05). Linear correlation analysis showed that CD34, PDGFR-β protein expression was not related (P> 0.05). Among 39 DFSPs, 9 were aneuploid (23%) and the others were diploid with nearly diploid aneuploidy (DI = 1.16 ± 0.14). No one of 30 DFAs Ploidy. Correlation analysis showed that the expression of PDGFR-β protein in aneuploid group was higher than that in diploid group (P <0.05). Conclusion CD34 and PDGFR-β are closely related to the occurrence and development of protuberant skin fibrosarcoma.
