目的:通过测定温肾壮骨复方对体外3种自由基(DPPH自由基、ABTS自由基、羟基自由基)的清除能力,探讨其抗氧化作用机制。方法:测定复方和其单味药对DPPH自由基、ABTS自由基清除率,筛选出抗氧化性较强的单味药;然后测定复方和其单味药对DPPH自由基、ABTS自由基、羟基自由基的清除率,计算其IC50值。采用MTT法测出复方和其单味药对乳腺癌骨转移细胞株MDA-MB-231BO的抑制率。结果:在初筛实验中,得出补骨脂和蛇床子是抗氧化性较强的两味单味药。在机制探讨实验中,得出温肾壮骨复方清除DPPH自由基能力和清除ABTS自由基能力与补骨脂相当,同时温肾壮骨复方及补骨脂也表现出较好的抗肿瘤性。结论:温肾壮骨复方具有一定的抗氧化和抗肿瘤性,表现出对DPPH自由基、ABTS自由基和羟基自由基清除的协同作用。
OBJECTIVE: To investigate the anti-oxidative mechanism of Wenshen Zhuanggu Compound on the scavenging ability of three kinds of free radicals (DPPH, ABTS and hydroxyl radical) in vitro. Methods: The DPPH free radical scavenging activity and ABTS free radical scavenging activity of compound and its single herbs were determined and single antioxidant with strong antioxidative activity was screened. Then DPPH free radical, ABTS free radical, hydroxyl group Free radical scavenging rate, calculate its IC50 value. MTT assay was used to determine the inhibitory rate of compound and single herbs on the MDA-MB-231BO breast cancer cell line. Results: In the preliminary screening experiments, it was concluded that psoralea and Cnidium were two strong single herb with strong antioxidative activity. In the mechanism of experimental study, Wenshen Zhuanggu compound radical scavenging DPPH free radical scavenging capacity and the ability of scavenging ABTS equivalent and psoralen, while Wenshen Zhuanggu compound and psoralen also showed good anti-tumor. Conclusion: Wenshen Zhuanggu decoction has some anti-oxidation and anti-tumor effects and shows the synergistic effect on the scavenging of DPPH radical, ABTS radical and hydroxyl radical.
