目的观察尿道括约肌各组织成分年龄变化,探讨与增龄相关的括约肌功能障碍的形态学基础。方法分别取6、12、24、32月龄雌性未生育Fisher大鼠28只,每组7只,尿道标本常规石蜡包埋切片,Masson’s Trichrome染色,光学显微镜观察并用计算机图像分析系统定量分析尿道括约肌各组织成分和空间形态的年龄变化。结果尿道横纹肌含量从12月龄开始减少;从24月龄开始尿道黏膜变薄、黏膜下血管数量减少、尿道嵴面积缩小、尿道腔最长径/最短径比值增大,胶原结缔组织含量增加;尿道背侧壁厚度变薄,尿道腔面积扩大仅见于32月龄组。尿道平滑肌和尿道总面积随年龄增加没有发生明显变化。结论随着年龄增长,雌性大鼠尿道横纹括约肌减少致关闭尿道的动力减弱,尿道黏膜变薄使密封效果减弱,尿道腔扩大使关闭距离增加等系列变化可能是增龄相关的尿道括约肌功能障碍性疾病的形态学基础。
Objective To observe the changes of urethral sphincter tissues in different age groups and explore the morphological basis of sphincter dysfunction associated with aging. Methods Totally 28 female Fisheries rats of 6, 12, 24, and 32 months old were randomly divided into 7 groups. The urethral specimens were routinely paraffin-embedded and stained with Masson’s Trichrome. The urethral sphincter was quantitatively analyzed by light microscope and computerized image analysis Age changes in composition and spatial pattern of each organization. Results The content of striated muscle in urethra decreased from 12 months old. From the age of 24 months, urethral mucosa became thinner, the number of submucosal blood vessels decreased, urethral crest area decreased and the ratio of longest diameter to shortest diameter of urethra increased, while collagen connective tissue content increased. The thickness of the urethral dorsal wall became thinner and the area of the urethral cavity enlarged only in the 32-month-old group. Urethral smooth muscle and urinary tract area with age did not change significantly. Conclusions With the increase of age, the reduction of urethral transverse sphincter in female rats results in weakened dynamic force in closing the urethra. The thinning of the urethra reduces the sealing effect and enlargement of the urethral cavity to increase the closing distance.
