为探讨两种价态 (2价和 3价 )锰化合物对 SY5 Y细胞的细胞毒性和引起细胞凋亡的作用 ,选择了多巴胺能神经细胞 SH- SY5 Y细胞作为受试系统 ,细胞活力测定结果显示 Mn2 +和 Mn3+可依时间和剂量依赖性而使细胞的存活率减少 ,但 Mn3+的细胞毒性作用高于 Mn2 + ,相差显微镜下观察 ,Mn2 +和 Mn3+刺激细胞 2 4 h后 ,细胞变圆 ,贴壁状态不好 ,HE染色得到相似的结果 ,在用流式细胞仪观察 Mn2 + 和 Mn3+ 的致凋亡作用中发现 ,不同价态锰均可引起 SH- SY5 Y细胞发生凋亡 ,但 Mn3+ 的致凋亡作用强于 Mn2 + 。
