随着年龄的增长,心血管系统鸟苷酸结合蛋白(简称G蛋白)耦联受体介导的反应性也发生了改变。本文用WesternBlot和核酸杂交技术分别观察了6,24月龄的Fischer344大鼠主动脉、心房、心室及脑组织中Gsα,Goα,Giα及其相应的mRNA的变化。结果显示,主动脉Gsα含4个带(38,42,45和52kD),其中42kD带在24月龄大鼠显著下降(P<0.05),Goα和Giα各为单一带(39kD和41kD),两者24月龄也有显著下降(P<0.05),但它们的mRNA水平未见显著改变;心房、心室中Gsα有三个带(52,45和42kD),Goα和Giα各为一个带,在24月龄大鼠三者均未见显著改变,但Gi2α和Gi3αmRNA的含量显著增加(P<0.05)。上述结果提示:Giα、Goα的表达水平下降可能是衰老血管组织受体反应性下降的分子基础之一。
With age, the cardiovascular system guanylate binding protein (G protein) coupled receptor-mediated reactivity has also changed. In this paper, the changes of Gsα, Goα, Giα and its mRNA in the aorta, atrium, heart and brain of Fischer344 rats aged 6-24 months were observed by Western blot and nucleic acid hybridization. The results showed that there were 4 bands (38, 42, 45 and 52kD) in aorta Gsα, of which 42kD band decreased significantly in 24-month-old rats (P <0.05) ), Both at 24 months of age (P <0.05), but their mRNA levels did not change significantly. There were three bands (52, 45 and 42 kD) of Gsα in the atria and ventricle, and one each of Goα and Giα There was no significant change in the three groups at 24 months old, but the contents of Gi2α and Gi3α mRNA increased significantly (P <0.05). The above results suggest that the decreased expression of Giα and Goα may be one of the molecular bases for the decline of receptor reactivity in senescent vascular tissues.
