目的建立紫花苜蓿中考迈斯托醇的含量测定方法。方法采用RP-HPLC法,使用Kromasil C18柱(250 mm×4.6 mm,5μm),流动相为甲醇-体积分数为0.04%磷酸溶液(体积比为60∶40),流速为1.0 mL.min-1,检测波长为345 nm,柱温为25℃。结果考迈斯托醇质量浓度在0.75~22.50mg.L-1内呈良好的线性关系(r=0.999 6,n=5),平均回收率为98.45%,RSD为2.2%(n=6)。结论RP-HPLC法适用于紫花苜蓿中考迈斯托醇含量的测定。
Objective To establish a method for the determination of medroxol in alfalfa. Methods The RP-HPLC method was used on a Kromasil C18 column (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of 1.0% methanol-0.04% phosphoric acid solution (volume ratio 60:40) , The detection wavelength was 345 nm and the column temperature was 25 ℃. Results There was a good linearity (r = 0.999 6, n = 5) for the concentration of Comastostol between 0.75 and 22.50 mg · L -1, with an average recovery of 98.45% and a RSD of 2.2% (n = 6) . Conclusion RP-HPLC method is suitable for the determination of medroxol in alfalfa.
