目的 :研究PUF家族基因[Pumilio1(Pum1)和Pumilio2(Pum2)]在雌性生育能力和生殖细胞减数分裂过程中的作用。方法:在Pum1基因位置携带lox P位点的Pum2基因全敲鼠的基础上,利用生殖细胞特异性表达的Cre-Vasa-Cre介导Pum1基因条件性敲除,以此获得Pum基因双敲小鼠模型Vasa-cre:Pum1F/FPum2-/-(VDKO)。同时,对Pum基因双敲小鼠进行生殖表型分析。结果:1交配实验结果表明,雌性VDKO小鼠的生育能力显著下降,但仍可以产生后代;2VDKO小鼠卵母细胞体外成熟过程不受影响。结论:Pum蛋白不影响小鼠卵子发生发育过程。
Objective: To study the role of PUF family genes [Pumilio1 (Pum1) and Pumilio2 (Pum2)] in female fertility and germ cell meiosis. Methods: Based on the knockout of Pum2 gene carrying lox P site at the position of Pum1 gene, conditional knockout of Pum1 gene by Cre-Vasa-Cre specifically expressed by germ cells was used to obtain double knockout Rat model Vasa-cre: Pum1F / FPum2 - / - (VDKO). At the same time, reproductive phenotype analysis of Pum double knockout mice. Results: 1 The results of mating experiment showed that fertility of female VDKO mice was significantly decreased, but still can produce offspring; 2VDKO mouse oocytes in vitro maturation was not affected. Conclusion: Pum protein does not affect the mouse egg development process.
