运用蛋白质组学方法建立残胃癌和癌旁正常胃黏膜组织(pH 3~pH 10)蛋白质表达谱,分析残胃癌和癌旁正常胃黏膜组织中蛋白质表达的差异。通过双向凝胶电泳(2-DE)分离残胃癌组织和癌旁正常胃黏膜组织的总蛋白,并采用凝胶染色和Powerlook 2100XL扫描仪扫描凝胶以获得残胃癌和癌旁正常胃黏膜组织蛋白质表达谱,最终将蛋白质表达谱图经ImageMaster2D Platinum软件加以分析,得到差异表达蛋白结果。通过比较分析3对残胃癌和癌旁正常胃黏膜组织(pH3~10)蛋白质表达谱,共发现108个差异表达的蛋白质斑点,显著差异的蛋白质斑点38个,其中仅在残胃癌组织中表达的蛋白质点3个,残胃癌组织中失表达的蛋白质点7个,15个点在残胃癌组织中表达上调,13个点在残胃癌组织中表达下调。残胃癌与癌旁胃黏膜组织之间存在着显著的蛋白质表达差异。利用蛋白质组技术寻找残胃癌与癌旁正常胃黏膜组织的差异表达蛋白,将为阐明残胃癌发病的分子机制及发现残胃癌的诊断蛋白奠定了基础。
Proteomic methods were used to establish the protein profiles of gastric cancer tissue and adjacent normal gastric mucosa (pH 3 ~ pH 10), and to analyze the differences in protein expression between gastric mucosa and normal gastric mucosa. The total protein of gastric cancer tissue and adjacent normal gastric mucosa was separated by two-dimensional gel electrophoresis (2-DE), and the gel was stained with gel and Powerlook 2100XL scanner to obtain the residual gastric cancer and adjacent normal gastric mucosal tissue protein Expression profile, the final protein expression profile by ImageMaster2D Platinum software to be analyzed to obtain differentially expressed protein results. A total of 108 differentially expressed protein spots were found in 38 pairs of gastric cancer tissues and 38 specimens of normal gastric mucosa adjacent to normal gastric mucosa (pH 3 ~ 10), 38 of which were significantly different 3 protein spots, 7 protein spots missing in gastric cancer tissue, 15 spots were up-regulated in gastric cancer tissue and 13 spots were down-regulated in gastric cancer tissue. There was a significant difference in protein expression between gastric mucosa and adjacent gastric mucosa. The use of proteomic technology to find differentially expressed proteins in gastric cancer and normal gastric mucosa will lay a foundation for clarifying the molecular mechanism of gastric cancer and finding the diagnostic protein for gastric cancer.
