目的:探讨卷柏水煎液对心肌缺血再灌注损伤(MI/R)的影响及作用机制。方法:KM雄性小鼠随机分为正常对照组、MI/R损伤模型组、卷柏水煎液低剂量+MI/R组、卷柏水煎液高剂量+MI/R组,卷柏水煎液灌胃给药5d后,腹腔注射垂体后叶素和硝酸甘油复制心肌缺血再灌注模型,同时记录不同时点的肢体Ⅱ导联心电图,连续观察80min后,取小鼠心肌组织制作病理切片以观察心肌损伤情况;并测定心肌组织超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量。结果:与正常组相比,模型组小鼠心电图J点发生明显偏移;镜下观察心肌组织病理损伤程度明显加重;SOD活性较正常组降低(23.52±7.21比33.02±4.13,P<0.05);MDA含量较正常组增加(9.80±3.07比5.86±1.04,P<0.05)。与模型组相比,卷柏水煎液预处理组心肌组织病理损伤程度减轻;高剂量组心肌组织SOD活性明显高于模型组(31.96±5.82比23.52±7.21,P<0.05),MDA含量明显降低(5.50±1.80比9.80±3.07,P<0.05);低剂量组SOD活性及MDA含量与模型组比较无显著差异。结论:卷柏水煎液能减轻心肌缺血再灌注损伤,其机制可能与其增加缺血再灌注心肌组织中SOD活性,降低MDA含量有关。
Objective: To investigate the effect of Decoction of Selaginella on myocardial ischemia reperfusion injury (MI / R) and its mechanism. Methods: KM male mice were randomly divided into normal control group, MI / R injury model group, low dose of decoction of Selaginella tricuspidata + MI / R group, high dose of decoction of Selaginella tricolor, MI / R group, After intragastric administration for 5 days, pituitrin and nitroglycerin were injected intraperitoneally to establish models of myocardial ischemia-reperfusion. At the same time, ECG of limbs II at different time points were recorded. After continuous observation for 80 minutes, the myocardial tissue of mice was taken for pathological examination In order to observe the myocardial injury. The activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) in myocardium were measured. Results: Compared with the normal group, the J point of the ECG in the model group shifted obviously. The pathological changes of myocardium were observed under microscope. The SOD activity in the model group was significantly lower than that in the normal group (23.52 ± 7.21 vs 33.02 ± 4.13, P <0.05) ; MDA content increased compared with the normal group (9.80 ± 3.07 vs 5.86 ± 1.04, P <0.05). Compared with the model group, the degree of myocardial injury in the pre-treatment group was significantly reduced; the activity of SOD in the high-dose group was significantly higher than that in the model group (31.96 ± 5.82 vs 23.52 ± 7.21, P <0.05) (5.50 ± 1.80 vs 9.80 ± 3.07, P <0.05). There was no significant difference between the low dose group and the model group in SOD activity and MDA content. Conclusion: Decoction of Decoction of Cyperus atheroscleroticis can relieve myocardial ischemia-reperfusion injury, which may be related to the increase of SOD activity and decrease of MDA content in myocardial tissue after ischemia-reperfusion.
