目的 探讨仿刺参糖胺聚糖(HGAG)对肺癌患者外周血体外细胞免疫功能影响.方法 选取健康人群40名(健康组)和肺癌患者30例(肺癌组)抗凝外周血4 mL,应用淋巴细胞分离液采用Ficoll密度梯度离心法分离PBMC,体外与不同浓度HGAG共培养24 h.流式细胞仪检测CD45 RA及CD45RO,以及CD1a、CD83分子的表达.结果 健康组和肺癌组CD45RA和CD45RO培养前表达差异有统计学意义(P<0.05),培养24 h后差异明显(P<0.001).培养后肺癌组CD45RA和CD45 RO在10和50 μg·mL-1间表达无差异(P>0.05),而健康组在50 μg·mL-1时CD45RO表达有差异性(P<0.001),CD45RA表达无差异性(P>0.05).健康组与肺癌组在与50μ g·mL-1HGAG培养前、后CD1a表达无差异性(P>0.05);CD83表达在肺癌组有差异性(P<0.001),健康组无差异(P>0.05).结论 HGAG在一定浓度范围内可以下调CD45RA分子表达和上调CD45RO分子表达,以及促进肺癌患者DC成熟,体外调节肺癌患者的细胞免疫功能.“,”Objective To investigate the effect of Glycosaminoglycan from Apostichopus Japonius on cellular immuno-regulation function for peripheral blood cells from lung cancer patients in vitro.Methods 40 health persons (health group) and 30 lung cancer patients (lung cancer group) were selected and taken anticoagulation peripheral blood 4 mL.The peripheral blood mononuclear cells (PBMC) were separated by Ficoll density gradient centrifugation using Lydroxypropylmethyl Cellulose,and to co-culture for 24 hours with different concentration HGAG in vitro.The express of CD45RA and CD45RO,CD1a and CD83 molecules were detected by flow cytometry.Results The express of CD45RA and CD45RO had statistical significance before cultivation (P<0.05),but the statistical significance was more obviously after cultivation 24 hours (P<0.001) in the two groups.There was no statistical difference in CD45RA and CD45RO between 10 and 50μ g · mL-1in lung cancer group(P>0.05),while the CD45RO was difference (P<0.001),but the CD45RA was not (P>0.05)at 50 μg · mL-1 in health group after cultivation.The express of CD1a had no significance in the two groups before and after cultivation with 50 μg · mL 1HGAG (P>0.05).The express of CD83 had significance before and after cultivation in lung cancer group (P<0.001),but it had no significance in health group(P>0.05).Conclusion HGAG could down regulation CD45RA and up regulation CD45RO expression,as well as promote DC mature and improve cellular immune function for lung cancer patients in vitro in a certain concentration.
