目的:探讨安胃汤对实验性慢性萎缩性胃炎大鼠模型的EGF、EGFR mRNA表达的影响。方法:采用甲基硝基亚硝基胍(MNNG)慢性萎缩性胃炎大鼠模型,48只清洁级健康雄性Wistar大鼠随机分为空白组、病理模型组、安胃汤组、胃复春对照组共4组。实时荧光定量PCR方法观察安胃汤对实验性慢性萎缩性胃炎大鼠模型胃黏膜EGF、EGFRmRNA表达的影响,对后者进行图像分析及统计学处理。结果:与病理模型组相比,安胃汤组和胃复春对照组均显著降低EGFmRNA/β-actin值(0.32±0.04 vs 0.64±0.08,0.42±0.05 vs 0.64±0.08,P<0.01),安胃汤组优于胃复春对照组(P<0.01)。与病理模型组相比,安胃汤组和胃复春对照组均显著降低EGFRmRNA/β-actin值(0.56±0.11vs 0.78±0.14,0.67±0.08 vs 0.78±0.14,P<0.01),安胃汤组优于胃复春对照组(P<0.05)。结论:安胃汤可能通过降低胃黏膜EGF、EGFR mRNA表达而起到治疗CAG作用。
Objective: To investigate the effect of AnWei Decoction on the expression of EGF and EGFR mRNA in rat model of chronic atrophic gastritis. Methods: Forty-eight healthy male Wistar rats were randomly divided into blank group, pathological model group, Anweitang group and Weifuchun control group by using methyl nitrosothioguanidine (MNNG) chronic atrophic gastritis rat model A total of 4 groups. Real-time fluorescent quantitative PCR method was used to observe the effect of AnWeiTang on gastric mucosal EGF and EGFR mRNA expression in experimental chronic atrophic gastritis model. The latter was analyzed by image analysis and statistical analysis. Results: Compared with the pathological model group, the values of EGFmRNA / β-actin in the Anwei decoction group and the Wei Fuchun decoction group were significantly lower (0.32 ± 0.04 vs 0.64 ± 0.08, 0.42 ± 0.05 vs 0.64 ± 0.08, P <0.01) Anwei Decoction group was better than Wei Fuchun control group (P <0.01). Compared with the pathological model group, the ANGT group and the Wei Fuchun control group decreased the expression of EGFRmRNA / β-actin significantly (0.56 ± 0.11vs 0.78 ± 0.14,0.67 ± 0.08 vs 0.78 ± 0.14, P <0.01) The soup group was better than the Wei Fuchun control group (P <0.05). Conclusion: AnWei Decoction may play a role in the treatment of CAG by decreasing the expression of EGF and EGFR mRNA in gastric mucosa.
