目的探讨MCPH1(microcephalin 1)基因过表达对人肺癌A549细胞迁移及侵袭能力的影响。方法将质粒pcDNA3.1(-)/MCPH1(实验组)和pcDNA3.1(-)(阴性对照组)分别转染人肺癌A549细胞,采用Real-time PCR及Western blot法分别检测转染48 h后的细胞内MCPH1的表达;采用细胞划痕试验及Transwell试验分别检测MCPH1过表达对细胞迁移及侵袭能力的影响。结果实验组A549细胞中MCPH1水平明显高于阴性对照组,差异有统计学意义(P均<0.05);实验组细胞迁移及侵袭能力较阴性对照组明显降低,差异有统计学意义(P均<0.05)。结论 MCPH1基因过表达可明显抑制A549细胞的迁移及侵袭能力。
Objective To investigate the effect of microcephalin 1 gene overexpression on migration and invasion of human lung cancer cell line A549. Methods The human lung adenocarcinoma A549 cells were transfected with pcDNA3.1 (-) / MCPH1 (experimental group) and pcDNA3.1 (-) negative control group respectively. Real-time PCR and Western blot were used to detect the expression levels of miR- After intracellular MCPH1 expression; using cell scratch assay and Transwell assay were detected MCPH1 overexpression on cell migration and invasion ability. Results The level of MCPH1 in experimental group was significantly higher than that in negative control group (all P <0.05). The ability of cell migration and invasion in experimental group was significantly lower than that in negative control group (P < 0.05). Conclusion MCPH1 gene overexpression can significantly inhibit the migration and invasion ability of A549 cells.
